Overview

Probe specification

  • EML4, 2p21, Red
  • EML4, 2p21, Green

The EML4 Breakapart probe consists of a green 406kb probe and a red 353kb probe, which are positioned on each side of the EML4 gene.

Probe information

The protein encoded by the echinoderm microtubule associated protein-like 4 (EML4) gene is involved in microtubule formation and stabilisation1.

A novel gene fusion of EML4 and Anaplastic Lymphoma Kinase (ALK) was recently identified in Non-Small Cell Lung Cancer (NSCLC).

The EML4-ALK fusion was shown to result from an inversion within chromosome 2p, inv(2)(p21p23), and was detected in approximately 7% of NSCLC cases2,3.

Heat pretreatment

  • Heat 50ml Tissue Pretreatment Solution (Reagent 1) in a porcelain wash jar or coplin jar immersed in a waterbath until it is either boiling or 98 - 100°C.
  • Boil slides for 30 minutes (Note: different incubation times may be required depending on tissue fixation. A 30-minute incubation is a recommended starting point).
  • Wash in PBS or dH2O at room temperature (RT) for 2 x 3 minutes.
